Ulrick, Couple of questions. I am no expert, but love the planning process. I am still a noob at this, only been modeling for about 10 years.
Are you planning point to point or loop?

As for space, I feel that docks are a great solution for a lot of industry traffic in a small area. Warehouses can be worked into the backdrop and piers provide great modeling potential.
I like how Verne utilizes his space with his Estrella & Sonora Grande layout. Great switch back and I love the mine in the corner.

I have read that the min. radius for the 2-4-4 is 22".
Might start a thread in the Layout design forum. I have read many of Rob Chant's posts and he seems very knowledgeable.

I feel your pain, those O scale buildings are huge, while building a company store my daughter asked if I was building a doll house. I switched to O scale narrow gauge from HO and it took a while to know how much space to leave for structures, basically I had to plan as if I had have the space and think in HO, for example, if I have 10' x 2' for space I figured that if I were still in HO I would have 5' x 1', then you will understand just how little space you really have. I found that I had to really strip my layout down to just a few things and do them well rather than try and cram in too much and not be happy with any of it.
They say those 2-4-4s will run on 22" but I think most agree they need much larger than that to look right.
You may want to consider having a hidden staging yard that has a 22" radius minimum curve and use #6 turnouts, keep the curves out of view so the 2-4-4 will not be seen and look bad. your could use the 2-4-4 and the 4-4-0 to run in and out of staging and get a small porter to switch on the visible tracks ( or convert the 2-4-4 into a 2-4-0 with a tender). The small locomotive could switch the industries and build up trains to send into staging using the "big" engines. you could have a nice resort style station (partial flat against the backdrop) and consider your layout the end of the line and have tourist coming on a passenger train out of staging (behind a big engine), the train could be turned and sent back into staging, that would give some excuse for a passenger train.
you might want to consider cassette staging since trains will be short. I used it on my HO layout and it worked well, just have a track go off layout and have temporary support to hold the cassette, that way you can use all your space for the layout and not have to reserve some for staging.
